Crystallization and preliminary X-ray crystallographic analysis of lipase from Pseudomonas cepacia

Abstract

Large crystals of lipase from Pseudomonas cepacia have been grown at room temperature from solutions containing 2-methyl-2,4-pentanediol and sodium citrate. They grow within two weeks to typical dimensions of 1·0 mm × 0·5 mm × 0·3 mm. The crystals belong to the monoclinic space group P21; with unit cell parameters a = 84·91 A ̊, b = 47·33 A ̊, c = 86·00 A ̊, and β = 116·09 °. And they diffract to about 1·6 Å upon exposure to synchroton X-rays. X-ray data have been collected to 2·2 Å Bragg spacing from a native crystal.
