Crystallization and preliminary X-ray crystallographic analysis of lipase from Pseudomonas cepacia

Abstract

Large crystals of lipase from Pseudomonas cepacia have been grown at room temperature from solutions containing 2-methyl-2,4-pentanediol and sodium citrate. They grow within two weeks to typical dimensions of 1.0 mm × 0.5 mm × 0.3 mm. The crystals belong to the monoclinic space group P21, with unit cell parameters a=84.91 Å, b=47.33 Å, c=86.00 Å, and β=116.09°. And they diffract to about 1.6 Å upon exposure to synchroton X-rays. X-ray data have been collected to 2.2 Å Bragg spacing from a native crystal.
