Development of effective immobilization method using modified glutaraldehyde for GL-7-ACA acylase

Abstract

The objectives of this study are to develop a suitable immobilization method for GL-7-ACA acylase, and to enhance the stability of GL-7-ACA acylase immobilized on silica gels. In this study, each step in the immobilization procedure was optimized individually to improve the efficiency of overall immobilization procedure of GL-7-ACA acylase. The optimal conditions of each step were determined as follows, pretreatment by 35% hydrogen peroxide, silanization by 3-APTES in acetone, crosslinking by using modified glutaraldehyde and 10 mg/ml as amount of GL-7-ACA acylase added in immobilization procedure. We also investigated the effect of sodium borohydride in order to increase the stability of the linkage between enzyme and support after immobilization. The activity of immobilized GL-7-ACA acylase treated with 2%(w/w) sodium borohydride was remained about 86% after 20 times of reuse.
