N2 - Liquid biopsy has been arising to substitute tissue biopsy which has several significant drawbacks. microRNAs (miRNAs) were used as the biomarkers for various diseases including cancers, but conventional diagnosis methods suffered from high cost, false-negative results, or low sensitivity. Here, we developed the signal amplification method for multiplexed miRNA diagnosis method using hydrogel microparticles by modifying hybridization chain reaction (HCR) to hydrogel microparticle-based assay. By doing so, we succeed to amplify fluorescent signal intensity (and thus sensitivity for miRNA) by dozens of times. We also demonstrated the difference between the normal and the patients in specific miRNA profile from urinary exosomal miRNAs.
