Abstract
Pre-concentration of liquid sample without pH change is essential for diagnosis in patient samples with low concentration virus or biological molecules. Target molecule's isoelectric point (PI) value and optimal pH should be stable in antibody-antigen reaction [1], to facilitate robust and accurate detection. Previously, pre-concentration using a buffer drain technique is developed for concentrating protein. Previous technique has a problem of changing pH of sample when utilizing electric field in the sample reservoir [2]. Here, we developed a simple micro-fluidic device, which can concentrate particle without changes in pH, based PDMS utilizing ion concentration polarization (ICP) by ion selective layer made of Nafion [3].
